Mesylate and other forms need different calculations
The MedChemExpress Ibutamoren mesylate page lists CAS 159752-10-0, formula C28H40N4O8S2 and molecular weight 624.77 g/mol. These describe the mesylate. They are not universal properties of any vial labelled MK-677. Before calculating, check the salt name, structure, quantity and certificate for the actual batch.
Salt mass includes the counterion. Consequently, 1 mg mesylate does not mean 1 mg free base. Conversion to free-base equivalents needs confirmed formulas for both forms and the certificate's basis for reporting content. Do not subtract a remembered “typical salt percentage” or use molecular weight from a different product card.
HPLC peak-area purity does not establish the absolute ibutamoren content of weighed material. For accurate quantitative work, separately check assay, water and residual solvents. If a vial contains a filler or prepared formulation, total contents are not ibutamoren mass. Record which data were used.
What is known about solvent?
For its mesylate, MedChemExpress lists water at ≥50 mg/mL and DMSO at 50 mg/mL. The DMSO entry specifies ultrasonication and fresh solvent. For water, ≥ means dissolution was established at that concentration and saturation is unknown. It is not a measured maximum solubility or a guarantee for another MK-677 form.
An aqueous buffer with salts and proteins is not equivalent to pure water. If the method permits an aqueous stock, it may reduce the need for an organic vehicle, but compatibility with the eventual medium still needs checking. Bacteriostatic water contains an additional component and is not an automatic substitute for laboratory water of specified quality. The preservative also enters the experimental sample.
A small preliminary solution
First allow a tightly sealed cold vial to reach room temperature to avoid condensation on the powder. Take a small portion if permitted by the method and weighing accuracy. Prepare compatible containers, the supported solvent and labels. The SDS and local laboratory procedure determine handling and protective equipment.
Add a measured solvent volume, mix gently and check for a remaining solid phase. Do not raise temperature or change pH arbitrarily to obtain clarity. If ultrasonication is needed, monitor heating and follow the supported method. Do not apply a DMSO-specific note to any other solvent without checking.
A calculation for confirmed mesylate
At molecular weight 624.77 g/mol, 1.00 mL of a 10.0 mM stock requires 6.2477 mg mesylate: 0.0100 mol/L × 0.00100 L × 624.77 g/mol. This is a calculation example, not a recommended concentration. Making 10.0 mg up to a final volume of 1.00 mL gives nominal mass concentration 10.0 mg/mL and approximately 16.01 mM.
The molar calculation is valid only for the correct form and appropriate content basis. Do not confuse final volume with added solvent volume. For quantitative methods, make up volume using the accepted procedure. Do not attempt accurate weighing below the balance's working range: change preparation scale or use a verified intermediate dilution.
Dilution and vehicle controls
Adding 2.00 µL of a 10.0 mM stock to a final volume of 2.00 mL gives 10.0 µM. For a stock in pure DMSO, the final DMSO fraction is 0.10%. An aqueous stock does not introduce DMSO through this addition. This does not make every aqueous solution equally compatible: account for salts, pH and preservatives too.
The vehicle control should reproduce the final solvent composition and every additive without ibutamoren. Across a concentration series, maintain the same vehicle fraction where required by the method. Check the mixture in the actual medium immediately after addition and at the end of incubation. A precipitate-free stock does not guarantee a precipitate-free dilution.
What can distort measurements?
Cloudiness or crystals make nominal added concentration an unreliable description of dissolved compound. Do not filter away precipitate without evaluating losses or call a mixed suspension a solution. Critical concentrations need an appropriate analytical method. A photograph is useful for records but does not replace quantitative analysis or identity confirmation.
In receptor or signalling assays, assess cell viability and possible sample effects on signal detection. Vehicle, osmolality or pH differences can produce effects without the intended mechanism. Do not compare mesylate and free-base mass concentrations as equivalent molar exposures. Even equal molarity should be compared under the same matrix and timing.
Refrigeration can be the correct condition for dry mesylate
The cited manufacturer specifies 4°C, sealed storage and protection from moisture for dry mesylate. This illustrates why “all dry materials need long-term freezer storage” is not universal. If the actual batch label specifies another temperature, follow it. Do not freeze material arbitrarily or replace its specification with general peptide advice.
Solutions require separate evidence for solvent, concentration, temperature, container and permitted freeze–thaw cycles. Catalogue periods for another batch do not establish stability of your aqueous stock or culture dilution. Single-experiment portions reduce reopening but do not establish shelf life. Keep preparation dates and temperature-excursion records.
Separately label whether concentration refers to mesylate, free base or nominal material. When transferring stock to another operator, include the calculation and batch certificate. On changing supplier, recheck form, solvent and specification even when the short name MK-677 and declared mass remain the same. This helps avoid an unnoticed exposure change between series.
Common questions
Can free-base molecular weight be used for mesylate? No. Does ≥50 mg/mL mean maximum solubility? No, that entry does not establish saturation. Must dry mesylate be frozen? No, the actual batch specification applies. Does clarity prove correct concentration? No: correct mass, form, final volume and appropriate method controls are needed.
