The largest published human study of MK-677 (ibutamoren), Nass and colleagues, Annals of Internal Medicine, 2008, ran for two years and enrolled 65 healthy adults aged 60 to 81. Participants took a fixed oral dose of 25 mg daily with no escalation: the dose was never raised across the whole follow-up period. Over that time the researchers recorded increased pulsatile growth hormone secretion and a statistically significant rise in fat-free mass, with tolerability rated as generally good. A fixed once-daily dose fits the molecule itself: unlike injectable peptide growth hormone secretagogues, MK-677 is orally active and does not need multiple doses through the day.
