Practical dosing and effect details
The largest published human study of MK-677 (ibutamoren), Nass and colleagues, Annals of Internal Medicine, 2008, ran for two years and enrolled 65 healthy adults aged 60 to 81. Participants in the MK-677 group took 25 mg orally once daily (in the first year the control group took placebo). The dose was never raised; in four participants it was lowered to 10 mg because of higher fasting glucose or joint pain. Over that time the researchers recorded increased pulsatile growth hormone secretion and a statistically significant rise in fat-free mass, with tolerability rated as generally good. Once-daily dosing fits the molecule itself: unlike injectable peptide growth hormone secretagogues, MK-677 is orally active and does not need multiple doses through the day. The 10–20 mg range in the table is below the dose used in the Nass trial. In an earlier study by Chapman and colleagues (1996), older adults took 2, 10 or 25 mg daily for up to 4 weeks, and growth hormone levels rose with the dose.
