MIF-1 is the short name for the tripeptide Pro-Leu-Gly-NH₂ (proline-leucine-glycinamide), also called melanostatin. What MIF-1 is, briefly
MIF-1 is the endogenous tripeptide Pro-Leu-Gly-NH₂. It was first described as a factor that inhibits release of melanocyte-stimulating hormone, hence the older name melanostatin. Where the interest came from
The first wave of work landed in the 1970s. In a clinical observation, intravenous Pro-Leu-Gly-NH₂ potentiated the effect of levodopa, then the standard therapy for parkinsonism (The Lancet, 1975). That was a signal that the tripeptide interacts with the dopamine system somehow, even though it is neither a dopamine precursor nor a classical receptor agonist.
A second wave in the 2000s explained why. Pro-Leu-Gly-NH₂ turned out to act as a positive allosteric modulator of the dopamine D2 receptor: it binds not where dopamine itself docks but alongside it, changing how the receptor responds to its own ligand (J. Med. Chem., 2009). That is a fundamentally different logic from a direct agonist, and it is what made the peptide a convenient tool for studying dopamine-receptor allostery.
Why this matters for a researcher
An allosteric modulator is useful where a direct agonist is too blunt. It does not switch the receptor on by itself, it only tunes its sensitivity, so in lab models it lets you separate the baseline response from the modulated one. That is why Pro-Leu-Gly-NH₂ and its synthetic derivatives remain a subject of chemistry and pharmacology work rather than a forgotten historical find.
