Thymosin alpha-1, also known as thymalfasin, is a short natural peptide first isolated from thymic tissue. The thymus is the organ where T cells mature, so it is no surprise that all the interest in this molecule revolves around the immune system.

What thymosin alpha-1 is, briefly

Thymosin alpha-1 is a 28-amino-acid peptide. It is described as an immunomodulator: it does not "boost immunity" in the advertising sense but influences the maturation and activation of certain immune cells. The mechanistic breakdown is in the thymosin alpha-1 monograph.

Where the interest came from

The peptide was isolated and sequenced back in 1977: the paper described the isolation and sequence analysis of the immunologically active thymosin alpha-1 (Proc. Natl. Acad. Sci., 1977). A body of studies of varying quality has accumulated since, and a recent review tries to organize it, separating what has been shown under controlled conditions from what remains preliminary (World J. Virol., 2020).

Why researchers find it interesting

A modulator of the immune response is a convenient tool for studying how finely the balance between activation and tolerance is tuned. Unlike a blunt stimulant, thymosin alpha-1 acts more like a regulator, so it is used in immunology work to study T-cell maturation and dendritic-cell function.
