Bacteriostatic Water: Handling, Reconstitution and Storage

Purpose

Bacteriostatic water is a multi-dose solvent for reconstituting lyophilised research substances in the laboratory. The 0.9% benzyl alcohol restrains microbial growth, allowing multiple withdrawals from a single vial across the preservative window (typically ~28 days) without loss of working-solution sterility.

Physical form

A clear, colourless aqueous solution with no sediment or suspended particles. 10 ml vial: Type I glass, bromobutyl stopper, aluminium crimp. Clarity and freedom from particles are the basic visual check before use — turbidity, flakes or a colour change mean the vial is discarded.

Reconstitution chemistry

Benzyl alcohol mixes freely with water, so the solvent behaves essentially as pure water when dissolving peptides. Add the solvent slowly, directing the stream against the glass wall rather than onto the lyophilisate — this lowers local shear and foaming. After addition, swirl the vial gently or leave it to dissolve on its own; shaking creates air bubbles and mechanical stress that can denature sensitive peptides. The working concentration is calculated as reagent mass divided by the solvent volume added (mg/ml).

Storage

Before reconstitution, a lyophilised reagent is most stable dry, under refrigeration or freezing, protected from light and moisture. Unopened bacteriostatic water is stored at room temperature, protected from direct light. After reconstitution the peptide solution is kept refrigerated; the preservative guards against microbial growth but does not stop chemical degradation of the active substance — hydrolysis, oxidation, aggregation.

Stability

The stability of a reconstituted reagent is governed by the peptide itself, not the solvent. Benzyl alcohol can interact with some proteins, shifting the equilibrium toward aggregation-prone forms, so for especially sensitive protein reagents preservative-free water (single draw) or other compatible buffers are considered as alternatives. Freeze-thaw cycling of the reconstituted solution should be minimised.

Lab handling

Wipe the work surface and the stopper with alcohol before every puncture; use sterile needles and single-use syringes. Record the opening date of each vial. Do not mix solvents from different lots in one vial. The material is research-use-only and is not intended for administration to humans or animals.

Applications

Typical research uses: reconstituting lyophilised peptides and proteins for in vitro experiments, preparing stock solutions, and diluting reagents in analytical protocols (HPLC, immunoassays) where repeated withdrawal from a single vial is needed.
