Check chemical form before calculating
For its PXL770, MedChemExpress lists CAS 1523493-53-9, formula C23H18ClNO3S and molecular weight 423.91 g/mol. Use these only when the batch label and certificate identify the same form. Chlorine in a molecular formula does not itself mean a hydrochloride salt. Do not add an assumed counterion to molecular weight without structural confirmation.
Keep the lot number, mass, receipt date, specification and identity document. A solvate, water or filler can alter the basis of a quantitative calculation. HPLC peak-area purity does not measure absolute PXL770 content in the whole powder. Accurate exposure needs an appropriate assay rather than an arbitrary correction using purity percentage.
Supported solubility is solvent-specific
The manufacturer page examined lists DMSO at 120 mg/mL, requiring ultrasonication and fresh DMSO. This does not establish equal solubility in water, buffer or culture medium. Do not use organic-stock concentration as evidence that the intended aqueous working concentration can be achieved without precipitation.
The most concentrated stock is not always preferable. Accurate transfer may require an impractically small volume, and dilution can create local supersaturation. Choose a verified concentration compatible with pipetting accuracy and acceptable vehicle fraction. The actual batch specification takes precedence over this example from another supplier's catalogue.
Preparing a trial portion
Allow a cold vial to reach room temperature while closed. Opening cold powder in humid air can introduce uncontrolled water. Prepare compatible containers and solvent of the required grade. DMSO is hygroscopic, so record solvent opening date and storage conditions alongside reagent details. Follow the SDS and local procedure.
Start with a small portion. Add a measured volume, mix and inspect for a solid phase. If ultrasonication is specified, monitor temperature to avoid uncontrolled heating. Document any change in pH or vehicle composition. Absence of visible particles after processing is only an initial check, not a certificate of stability.
A molar stock example
For the confirmed form with molecular weight 423.91 g/mol, 1.00 mL of a 10.0 mM stock requires 4.2391 mg PXL770. Calculate mass = molar concentration × final volume × molecular weight. Another example: 5.00 mg in a final volume of 1.00 mL gives nominally 5.00 mg/mL, approximately 11.79 mM.
These are arithmetic examples, not recommended experimental concentrations. They assume correct identity and an appropriate quantitative-content basis. Use final solution volume rather than simply added solvent volume. Do not claim better weighing precision than the balance provides; change preparation scale or prepare a verified intermediate dilution when necessary.
How much DMSO enters the experiment?
Adding 2.00 µL of a 10.0 mM stock to a final volume of 1.00 mL gives 20.0 µM PXL770. For stock in 100% DMSO, this also gives 0.20% DMSO by volume. For a stock containing 50% DMSO, the result is 0.10%. Sum DMSO contributions from every reagent if there are several.
Cells, duration and endpoint determine the acceptable fraction. Do not automatically consider an organic vehicle neutral when measuring metabolism. The vehicle control reproduces the final composition without PXL770. Maintain a consistent vehicle composition across concentration series. Otherwise changes in oxygen consumption, ATP or viability may partly reflect DMSO differences rather than the compound.
Check the aqueous dilution separately
Add a trial stock portion to the actual medium under the main experiment's conditions. Inspect immediately and after the required interval. Protein, salts, pH and temperature can change behaviour compared with pure water. A clear stock before addition does not establish suitability in medium. Separately assess the highest intended working concentration.
If precipitate appears, nominal added amount is not a measured dissolved concentration. Filtration may remove compound rather than fix preparation. Do not return a suspension to use without reconsidering the method. For quantitative comparison between series, use a validated analytical assessment of actual available material and record any procedural change.
AMPK and energy-metabolism assays
A change in cellular ATP or protein phosphorylation does not alone establish a direct mechanism. Check viability, vehicle and protocol-specific endpoints. In a cell-free enzyme assay, record enzyme-complex composition, substrate and ATP. Do not treat different assay results as equivalent simply because they use the same PXL770 concentration in µM.
If detection uses luminescence, fluorescence or coupled enzymes, include an appropriate interference control. An analyte or vehicle can affect measurement independently of the cellular pathway. This is not a claim that PXL770 necessarily produces such interference; the check separates biology from properties of the particular detection system.
Dry PXL770: do not import a freezer rule
The cited manufacturer specifies 4°C, sealed storage and protection from moisture and light for dry material. Automatic long-term freezing of all reagents therefore does not follow from this source. The actual vial's label governs its storage. If a separate specification permits long-term freezer or short-term refrigerator conditions, record that basis rather than an analogy with peptides.
Solutions have separate requirements. Label composition, concentration, date, container and freeze–thaw cycles. Do not transfer organic-stock catalogue periods to an aqueous working dilution, another solvent or another batch. Single-experiment portions reduce reopening but do not prove stability. Critical work using stored stock requires comparison with fresh material.
Common questions
Can preparation start with bacteriostatic water? The cited data do not support that. Should HCl be added to a formula containing Cl? No: structure and certificate establish form. Can DMSO below one percent be ignored? No, compatibility is method-dependent. Does clarity after dilution establish full stability? No, visual inspection does not replace chemical checks.
