β-NMN: identity before dissolution
β-Nicotinamide mononucleotide, or β-NMN, is a phosphorylated precursor of NAD⁺. It differs from nicotinamide riboside, NR, by a phosphate group, and from nicotinamide by its chemical structure. The description “NAD precursor” does not make these substances interchangeable by mass, molar concentration or behavior in cell culture medium. The NR → NMN → NAD⁺ pathway and its limitations are discussed in the NR/NMN review.
Check the label and COA for the β-form specifically, CAS, formula weight, hydration state and any salts or excipients. One documented example is Cayman Chemical No. 16411, β-NMN, CAS 1094-61-7, C₁₁H₁₅N₂O₈P, formula weight 334.2 g/mol. This is not the molecular weight of NR chloride or a universal conversion factor for every product labelled NMN. If the material contains another salt or water, the calculation needs information for that particular form.
What needs to be known about the batch
Record LOT, receipt date, material mass, transport conditions and labelled expiry. Distinguish HPLC peak-area purity from quantitative β-NMN content. The former describes signal distribution between detected peaks but does not by itself account for all water, counterions or components invisible to the detector. Do not multiply the vial mass by an arbitrary “purity percentage” unless the COA defines it as a mass fraction appropriate for that calculation.
For cell experiments, assess microbiological suitability and endotoxins separately where relevant to the model. Analytical calibration requires a standard with assigned content and traceable preparation. A commercial research reagent does not become a certified standard simply because its HPLC value is high. Resolve discrepancies in the name, formula or LOT before opening, while the original material has not yet been mixed with vehicle.
Aqueous stocks and matrix compatibility
The Cayman specification describes β-NMN as a crystalline solid that can be dissolved directly in aqueous buffers. It reports approximate solubility of 10 mg/ml in PBS at pH 7.2. This is useful for checking whether a trial calculation is plausible, rather than a guaranteed limit for another salt, temperature or concentrated mixture. For an ordinary aqueous experiment, do not add an organic vehicle without a reason and a control for its effects.
In LC-MS, the vehicle must suit the validated method: PBS introduces nonvolatile salts that can suppress ionization. For cell samples, account for final medium pH and osmolality, particularly when adding a large proportion of concentrated stock. If sterile filtration is needed, check β-NMN recovery across the membrane. Filtration neither removes all endotoxins nor distinguishes original NMN from its soluble conversion products.
Preparation sequence
Keep the chilled dry vial closed until its temperature equilibrates to avoid collecting condensation. Take a small trial amount, add part of the calculated vehicle, mix until dissolved and bring to the final volume. Record concentration, buffer composition, pH and preparation time. Clarity is a useful visual check, but it does not confirm chemical identity or exact β-NMN content.
Do not use heating, strong acid or alkali just because the powder dissolves more slowly than expected. Check the actual mass, chemical form and chosen vehicle. If the method requires another pH, first assess the change in a separate sample and check its analytical profile. Keep the vehicle selection and pH adjustment history with the results; otherwise, two nominally identical concentrations may represent different experimental conditions.
Calculating molarity and dilution
For a verified form with a formula weight of 334.2 g/mol, 3.342 mg in a final volume of 1.00 ml gives 10.00 mM: 0.003342 g ÷ 334.2 g/mol ÷ 0.001 l. That stock has a mass concentration of 3.342 mg/ml. The example assumes 100% content of the stated compound and complete dissolution. Correct the calculation when an assigned mass fraction is available; do not assign an exact molarity to an unidentified formulation.
For 1.00 ml of a 0.20 mM working solution from a 10 mM stock, take 20 µl and add vehicle to 1000 µl. If that solution is subsequently diluted in medium, include the next step using C₁V₁ = C₂V₂. For example, 100 µl of this solution plus 900 µl of NMN-free medium gives 0.020 mM, or 20 µM. This is laboratory dilution arithmetic, not a selection of a biologically effective concentration or a human administration schedule.
From stock solution to a biological result
An increase in cellular NAD⁺ after adding NMN does not prove that all added material remained NMN or entered the cell directly. The review by Yoshino, Baur and Imai discusses precursor conversion, including possible extracellular conversion of NMN to NR. Medium composition, serum, incubation time and matrix enzyme activity therefore affect interpretation. Starting concentration in a tube and cellular exposure are different measurements.
Include a vehicle control, fresh β-NMN and a cell-free medium control subjected to the same incubation period. Where possible, compare NMN, NR and nicotinamide profiles at the start and end of the experiment using a method with verified separation. For LC-MS, assess standard addition, matrix effects and injection carryover. The UV signal near 265 nm reported by the manufacturer is not specific proof of unchanged β-NMN in a complex medium.
Dry storage, aliquots and working time
Cayman specifies −20 °C for its dry product No. 16411. Do not automatically transfer that temperature to every NMN form: conditions for the Longeva batch must be supported by its documentation. Keep the dry material tightly closed with minimal moisture entry. Do not use a weighed portion with an unknown open-vial history as a precise calibrator without checking content again.
Cayman does not recommend keeping its aqueous solution longer than one day. Prepare enough for the current run; refrigeration alone does not establish a long shelf life. Freezing and a longer period require a separate verified protocol for the same matrix and concentration. If that protocol specifies aliquots, make single-use portions and record thawing. Compare stored stock with fresh stock by content and impurity profile, rather than cellular effect alone.
Records and sources
The record should connect LOT, exact form, calculation, vehicle, date and time, temperature, aliquot and control result. New cloudiness, precipitation, disagreement with a standard or an unknown temperature history requires assessment before continuing. Check information against the β-NMN product page, COA archive and NR chloride guide when comparing precursors.
