NR and NR chloride: what is on the label?
Nicotinamide riboside, NR, is a precursor of NAD⁺. In the pathway involving nicotinamide riboside kinase, NR is converted first to NMN and then to NAD⁺. Laboratory catalogues often supply it as the chloride salt, NR-Cl. The abbreviation NR describes the biological component, but weighing requires the full salt composition. Do not use the molecular weight of NMN, nicotinamide or another NR salt simply because all these materials participate in NAD metabolism.
MedChemExpress lists formula C₁₁H₁₅ClN₂O₅ and molecular weight 290.70 g/mol for nicotinamide riboside chloride HY-123033A. The molar examples below apply specifically to verified NR chloride with that weight. They do not establish the composition of another manufacturer’s vial. Check the form, hydration state, LOT and quantitative content in your batch COA. Biological context is available in the NR monograph.
Moisture, content and weighing accuracy
The manufacturer of HY-123033A requires protection of dry material from moisture and light. In practice, open the dry vial briefly, use clean tools and close it tightly after sampling. Let chilled packaging reach room temperature while closed before opening. Condensation or absorbed water changes the weighed mass even when the main component’s chromatogram appears acceptable.
Do not equate HPLC peak-area purity with the mass fraction of NR chloride. An accurate standard requires assigned content with a defined treatment of water and salt. If this is unavailable, record that the concentration was calculated from nominal mass. For cell studies, separately check sterility or microbiological suitability and endotoxins when the model requires them. Neither the name NR-Cl nor a high HPLC percentage proves those characteristics.
Choosing an aqueous vehicle
MedChemExpress reports water solubility of 125 mg/ml for its NR chloride, with a note that sonication is required. This is a result for that product and the manufacturer’s conditions, not a requirement to prepare such a concentrated stock. For small trials, first check the lower concentration actually needed in the chosen vehicle. Do not transfer the sonication note to every dilution; if the protocol requires it, monitor sample heating.
Water, PBS and complete culture medium are different matrices. In an enzyme experiment, match pH and ionic strength to the reaction. For LC-MS, avoid unplanned nonvolatile salts and check ion suppression. DMSO is not mandatory simply because the manufacturer also lists solubility in it; an organic vehicle introduces another variable for cells and analysis. Bacteriostatic water likewise requires assessment of preservative effects rather than automatic selection as a universal solvent.
A trial before the full run
Prepare a small weighed portion, add part of the final volume of water or compatible buffer and mix. After dissolution, bring to the required final volume. Record pH, vehicle composition, temperature and time. If the powder does not dissolve under expected conditions, check form, concentration and documentation before changing pH or heating. Strong acid or alkali is not a chemically neutral way of “helping” the reagent.
When filtration is needed, check membrane material and NR recovery after passage. Compare samples before and after filtration using the same method. Absence of particles after a filter does not confirm unchanged chemical composition, and a sterile membrane does not establish absence of endotoxins. For an instrumental run, also check vessel compatibility, background signal and carryover from previous injections.
A calculation for the chloride salt
With a verified NR-Cl molecular weight of 290.70 g/mol, 2.907 mg in a final volume of 1.00 ml gives 10.00 mM: 0.002907 g ÷ 290.70 g/mol ÷ 0.001 l = 0.01000 mol/l. The mass concentration is 2.907 mg/ml. This assumes that the weighed portion consists entirely of the specified salt and dissolves completely. Another salt or assigned content below 100% needs a different calculation; peak area alone does not provide that correction.
For 2.00 ml of a 0.10 mM working solution from a 10 mM stock, take 20 µl of stock and add vehicle to a final 2000 µl. Check: 10 × 20 = 0.10 × 2000 when concentrations are in mM and volumes in µl. Subsequently mixing 100 µl of working solution with 100 µl of NR-free medium gives 0.050 mM in the experiment. These numbers illustrate arithmetic; choose the biological concentration for the experimental question, rather than dilution convenience.
What happens in the medium
The study by Ratajczak and colleagues demonstrates the role of NRK1 in NR and NMN metabolism in the cell models studied. It does not establish an identical response in every cell line. NR can be converted within a biological matrix, so final cellular NAD⁺ is not a direct measurement of unchanged NR in the medium. Account separately for serum, basal medium composition, cell density and incubation time.
Include a vehicle control, cell-free medium containing NR, a fresh NR control and, when comparing pathways, a separate nicotinamide control. Chemical analysis requires separation of NR, NMN and nicotinamide; a nonspecific UV signal alone is insufficient. Check recovery of a standard added to the matrix and stability while queued in the autosampler. For a long experiment, compare composition at the start and end instead of attributing the entire effect to unchanged NR.
Storage without a universal rule
For dry HY-123033A, MedChemExpress specifies 4 °C, sealed storage protected from moisture and light. This is a concrete reason why “put every research powder in the freezer” is unsuitable here. It does not replace the Longeva batch label. If that specification gives different conditions, follow the verified requirements for that form. Also establish whether the recommendation applies to unopened or previously opened packaging.
The MCE page lists separate conditions for material “in solvent,” including −80 and −20 °C. Do not transfer a general catalogue period to every aqueous buffer, culture or diluted working sample: composition and concentration must match the verified protocol. Until such a period is established, prepare a fresh quantity for the run. If a documented procedure allows freezing, divide the stock into single-use aliquots and avoid repeated thawing.
Records and sources
For each stock, retain LOT, salt form, calculation basis, concentration, pH, vehicle, date and time, storage conditions and control result. Precipitation, profile changes, loss of content or unknown temperature history needs investigation. Plan NR and NMN comparisons using their different molecular weights, with the β-NMN guide, NR-Cl product page and batch COA.
