Nicotinamide mononucleotide (NMN) is a direct precursor of NAD⁺, the coenzyme that fuels mitochondrial respiration, sirtuin activity, and DNA-repair enzymes. In the canonical salvage pathway, NAMPT converts nicotinamide into NMN, and the NMNAT enzyme family (NMNAT1–3, each anchored in a different cellular compartment) completes the final step to NAD⁺ using ATP. That puts NMN one enzymatic step closer to the finished coenzyme than NR: nicotinamide riboside first needs phosphorylation by the NRK1/NRK2 kinases before it reaches the same NMNAT step.
How extracellular NMN actually gets inside a cell is still debated. A 2019 paper described Slc12a8 as a dedicated NMN transporter in mice; later independent work could not reproduce the finding and questioned it. A competing line of evidence holds that extracellular NMN is first dephosphorylated to NR by the ecto-enzyme CD73, crosses the membrane through the ENT1/ENT2/ENT4 nucleoside transporters in that form, and is rephosphorylated back to NMN once inside. Which route dominates in humans is not yet settled.
In human trials, oral NMN reliably and reproducibly raises blood NAD⁺ — that part is consistent across studies. The functional picture is messier: a systematic review and meta-analysis of randomised trials (342 participants, doses of 250-2000 mg/day) found no statistically significant effect on fasting glucose, insulin, or lipid profile overall, while a separate 10-week trial in prediabetic postmenopausal women reported improved insulin-stimulated glucose uptake in skeletal muscle. Short-term trials have not reported serious adverse effects.
Unlike NR, which carries GRAS status in the US and novel-food authorisation (as the chloride salt) in the EU since 2020, NMN's US regulatory path has been rockier: in late 2022 the FDA determined NMN was excluded from the dietary-supplement definition, citing its earlier authorisation for drug investigation (an IND exclusion). Following a petition and lawsuit from industry groups, the FDA reversed that position in 2025. Inside this catalogue that history is background context, not a usage cue — every item here is sold strictly for laboratory and research use, not for human consumption.
