Practical dosing and effect details
In the two randomized RECONNECT phase 3 trials (Kingsberg and colleagues, Obstetrics & Gynecology, 2019), 1.75 mg subcutaneous bremelanotide given on demand was compared with placebo. Participants took the dose roughly 45 minutes before sexual activity rather than on a fixed daily schedule.
In that trial it was this single on-demand dose, not a course with gradual escalation, that showed a statistically significant difference from placebo on the primary endpoints (desire and distress). That sets bremelanotide apart from the daily- or weekly-schedule peptides elsewhere on this site: the effect here is tied to one administration, not to a dose accumulated over a week.
