Full-length Tβ4 contains 43 residues

Thymosin β4 is a naturally occurring, 43-residue N-terminally acetylated peptide. Full-length Tβ4 and a short fragment sold as TB-500 cannot be treated as the same reagent. The TB-500 trade name alone does not guarantee length because commercial usage varies. This page requires a confirmed full Tβ4 sequence, terminal modifications and expected mass. Establish whether the supplied material is the parent peptide, a labeled probe or a mixture. An internal short motif does not automatically reproduce every property of the entire peptide.

Batch identity and supplied form

Before opening, match the vial name, batch number, certificate of analysis and specification. Establish identity, stated mass, counterion, water, excipients and recommended conditions. “Lyophilized” describes a drying process; it does not establish purity, sterility or exact peptide content. HPLC main-peak area and peptide mass fraction are different measurements: water and salts may contribute little to that chromatogram. If the manufacturer has not established peptide content, label the concentration as calculated from nominal mass.

DNase I and polymerization are different assays

The 1993 primary study assessed Tβ4 actin-sequestering properties through DNase I inhibition: G-actin inhibits the enzyme whereas F-actin does not. Full-length peptide and fragments were compared; truncation did not reproduce every full-length function. Establish whether your quality-control assay measures available G-actin, filament formation or the complex itself. These are related but different quantities. An additive affecting DNase I or its substrate can change the enzymatic endpoint without the intended actin change, requiring reaction-component controls.

Actin experiments require stoichiometry

Full-length Tβ4 binds monomeric G-actin; cell-free experiments depend on both partner concentrations and the initial actin state. Fluorescent labeled-actin polymerization assays need a no-peptide control, peptide-background control and matched salts and nucleotides. Decreased fluorescence alone does not prove specific binding: optical interference, precipitation or an altered actin state can contribute. For stoichiometric comparisons, establish Tβ4 concentration from its confirmed full-length form rather than a TB-500 fragment mass.

Mass calculations and working dilutions

An arithmetic example, not a solubility prescription: 2 mg in a final volume of 4 mL gives 0.5 mg/mL, or 500 micrograms/mL. Adding 20 microliters of that stock to 980 microliters of compatible matrix gives 10 micrograms/mL. Then 100 microliters of this solution plus 900 microliters of matrix gives 1 microgram/mL. Use the final volume in the calculation. If the certificate separately establishes peptide mass fraction, apply it according to the labeling convention, without correcting the mass twice. Molarity requires the molecular weight of the confirmed form; do not borrow a value from a related analogue.

Methionine and matrix matter

Tβ4 contains methionine; oxidation can produce a distinct chemical form that should not be pooled with the parent material without assessment. Control oxidant, light and air exposure according to the method. This does not justify adding a reductant to an actin assay without testing: the additive can affect the partner protein and fluorescent probe. Check the solution after transfer to polymerization buffer separately. A clear aqueous stock does not establish quantitative recovery in a medium with different ionic strength.

Oxidation does not necessarily abolish every function

Oxidized Tβ4 retained actin-sequestering ability in the 1993 study. A 1995 study using equilibrium centrifugation, cross-linking and viscometry showed altered interactions after Met6 oxidation and N-terminal truncation, with effects dependent on form and endpoint. A new oxidized form therefore implies neither automatically zero activity nor unchanged activity. Batch control needs both parent-material fraction and the chosen functional response. “Activity retained” without an identified method conceals that distinction.

Functional suitability questions

Can a migration assay replace actin-binding analysis? Migration depends on many processes and is not a direct stoichiometric measurement. Is an unchanged HPLC peak sufficient? Assess mass and impurities too, plus the relevant actin function. Can TB-500 instructions be used? Only after confirming the same full-length molecule, salt form and formulation. Compare stored material with fresh Tβ4 and verified actin in the same series to separate peptide failure from a problem with its protein partner.

Actin source and reagent storage

The 1995 work compared actin sources including bovine heart and rabbit skeletal muscle. The partner protein also defines the experiment. Record its source, form, preparation and initial state. Store dry Tβ4 sealed and frozen as specified; establish actin conditions separately. Do not freeze a mixture according to only one component’s instructions. When testing peptide storage, add it to freshly checked actin to separate peptide changes from partner-protein aging or shifts in the G-actin/F-actin balance.
