Native humanin and its HNG (S14G) analog act on the same signaling pathway, but a single point substitution of serine for glycine at position 14, hence the abbreviation S14G, raises the analog's affinity for the receptor complex. That is why the schedule here doses HNG at roughly a tenth of the native peptide's amount (100-500 mcg versus 1-3 mg) at the same frequency, 2-3 times a week: higher affinity means less material is needed to produce the same signal. Sources administer both forms subcutaneously in short cycles (4-8 weeks) with a break, rather than continuously: controlled human clinical trials of humanin are scarce, and the published regimens lean mainly on preclinical work and community research protocols rather than an approved clinical schedule.
